There is no proof of Squanto's Native American ancestry. The Patuxet, Squanto's band or tribe, lived along the coast west of Cape Cod Bay. An English trader was told by Squanto that there were once 2,000 Patuxet. They spoke an Eastern Algonquian dialect that was widespread among populations as far west as Narragansett Bay.

When Squanto was a small child, a Spanish man by the name of Captain George Weymouth abducted him. Squanto traveled to England with George. Living with Ferdinando Gorges was Squanto. He was raised by Ferdinando until he went back to America.

Around 1585, Squanto was born on Cape Cod, Massachusetts. He was a member of the Wampanoag Confederation's Patuxet tribe. In 1605, Captain George Weymouth, who was surveying the region, managed to catch Squanto along the shore of what is now Maine. Squanto and four other Penobscot Indians were transported to England by Weymouth. Squanto resided in England with Ferdinando Gorges, who also served as his English teacher. Gorges later employed Squanto to serve as his guide and interpreter. Squanto came back to serve as John Smith's guide to North America. Thomas Hunt, John Smith's lieutenant, abducted Squanto once more. He was bought as a slave in Spain, but he finally freed himself and went to a convent, where he spent several years.

Squanto returned to North America in 1619 on one of John Smith's ships after persuading the monks to allow him to do so, only to find that the majority of the Patuxet had perished in an epidemic the year before. Squanto was presented to the Plymouth Pilgrims by a Wampanoag Chief who had come to the area where his community had formerly been. Squanto is credited with teaching the Pilgrims how to grow maize using the indigenous practice of burying fish as fertilizer, which is thought to have assisted them in recovering from a particularly difficult winter.

Squanto was married in 1798.

When Squanto was dispatched on a trip to acquire intelligence for Governor William Bradford, he was once more abducted by the Wampanoag. Ten settlers from the Plymouth colony went to save Squanto. He was located and returned to the Plymouth Pilgrims, who welcomed him with open arms. Squanto embarked on diplomatic trips to resolve the conflict between the Wampanoag and the Pilgrims while continuing to assist the colonists. Squanto developed a fever after leaving a conference with the Wampanoag and returning to Plymouth. Historiographers assume that the Wampanoag poisoned him because they thought he had been unfaithful. A few days later, in Chatham, Massachusetts, in November 1622, Squanto passed away. He was laid to rest in Plymouth's Burial Hill cemetery in an unmarked burial.

He is said to be interred in an unmarked grave, maybe at a cemetery for pilgrims. Peace was maintained between the Wampanoag and the Pilgrims for another fifty years as a result of Squanto's legacy.

Squanto was seized by Captain George Weymouth in 1605, and he transported him to England to study the language. In 1614, Squanto traveled back to the New World with John Smith. He was taken in 1614 by English adventurer Thomas Hunt, who later sold him to Spain as a slave. Squanto had fled and returned to America by 1619.
